This Human Angiopoietin-Related Protein 6 ELISA Kit is intended for quantitative detection of human ANGPTL6 in cell culture supernates, serum and plasma (heparin, EDTA). Strip well format. Reagents for up to 96 tests.
This human ANGPTL6 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for ANGPTL6 has been precoated onto 96-well plates. Standards (Expression system for standard: CHO, Immunogen sequence: Ala20-Leu470) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for ANGPTL6 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substmousee TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Fibronectin amount of sample captured in plate.

Expression system for standard: Angiopoietin-like 6 (ANGPTL6), also known as angiopoietin-related growth factor (AGF), is a secreted 50 kDa protein that contains a coiled-coil domain and a fibrinogen-like domain. It is mapped to 19p13.2. ANGPTL6 is highly expressed in hepatocytes and is detectable in serum. Within the liver, secreted ANGPTL6 inhibits gluconeogenesis and promotes insulin sensitivity and energy expenditure. Circulating ANGPTL6 promotes the chemotaxis of vascular endothelial cells resulting in increased vascular permeability and angiogenesis. An integrin--binding RGD motif located within the fibrinogen--like domain of ANGPTL6, enables it to promote skin wound healing by mediating the adhesion and migration of keratinocytes, fibroblasts, and endothelial cells. Elevated levels of ANGPTL6 in serum are associated with metabolic syndrome, diabetes, and pre-eclampsia. Serum ANGPTL6 levels are also elevated during obesity. ANGPTL6 is reported to promote the survival and expansion of hematopoietic stem cells. ANGPTL6 is produced by several hematopoietic cell types including megakaryocytes, platelets, mast cells, and uterine NK cells.
